Construction of an optical system (Practical): Single-molecule imaging using a total internal reflection fluorescence microscope (TIRFM)
Single-molecule imaging of fluorescent dye in an aqueous environment with a total-internal reflection fluorescence microscope is explained. Specific topics include; spatial resolution, localization precision, detailed examples and tips for imaging and data analysis. We will also introduce high-speed, high-localization precision single-molecule imaging with a total-internal reflection dark-field microscope and gold nanoparticles as a probe.
